What is AAV?

AAV is the acronym of Adeno-Associated Virus, a non-enveloped virus with a single stranded DNA genome. The recombinant AAV (rAAV) has become popular as an efficient vehicle to mediate DNA delivery into mammalian cells, both in vitro and in vivo.

How to use AAV particles to deliver a piece DNA?

  1. Clone the transgene into an AAV vector, between 2 ITR sequences.
  2. Prepare auxiliary plasmids encoding key AAV proteins, including serotype-determining Rep-Cap
  3. Co-transfect above mentioned plasmids at optimal level into packaging cells
  4. Harvest and concentrate the AAV virus

What are AAV serotypes and how to select?

One of the useful features of AAV is its serotypes, which defines the preference of the types of cells/organs they infect. The tables below provide the selection guide for in vitro and in vivo applications.
