CRISPR RNP Delivery Platform

ProteanFect™ CRISPR RNP
Transfection Kits

A coacervate-based, protein-engineered delivery system that gets Cas9 RNP complexes into the primary and hard-to-transfect cells that matter — with no electroporator, no virus, and no lipid-driven toxicity.

Why ProteanFect

Built for the cells conventional transfection can't reach

Lipofection stresses primary cells. Electroporation demands dedicated hardware and a BSL-2 upgrade. ProteanFect's engineered-protein coacervate mechanism was designed to solve both problems at once.

High efficiency in cells that matter

Up to 88.9% mRNA transfection in primary human T cells, and 83% indel efficiency in primary mouse T cells — validated data, not estimates.

No specialized equipment

Works with standard cell culture equipment and serum-free media. No electroporator, no viral production facility, no BSL-2 upgrade required.

Cell viability preserved

The gentle coacervate mechanism maintains cell health. Viability is typically restored by day 2 — cells remain functional for flow cytometry, sequencing, and adoptive transfer.

The Technology

Coacervate-based RNP delivery mechanism

Engineered mammalian proteins (Reagents A, B, C) encapsulate Cas9 RNP into a stable coacervate complex, deliver it into the cytosol, and step out of the way.

1

Coacervate Formation

Engineered mammalian proteins (Reagents A, B, C) encapsulate the RNP into a stable coacervate complex.

2

Cellular Uptake

Coacervates undergo efficient cellular internalization, followed by robust cytosolic release of bioactive cargo.

3

Genome Editing

Cas9 edits the target locus, then degrades — limiting off-target nuclease activity versus plasmid or viral expression.

Validated Performance

TRAC knockout in primary T cells, quantified two ways

Positive-control sgRNAs targeting the TRAC/Trac locus were used to benchmark editing at 72 hours post-transfection by flow cytometry (CD3–TCR protein loss) and TIDE analysis of Sanger sequencing (genomic indel rate).

Simplified Workflow

Complex to result in under 3 hours

Simplified transfection protocol shown per well of a 96-well plate. Scales linearly to 48-, 24-, 12-, and 6-well formats.

1

Complex Preparation

Mix 0.8 µg Cas9 + 0.3 µg sgRNA with 40 µL Reagent A. Add Reagent B, mix thoroughly, then add Reagent C and place on ice. Use within 30 minutes.

2

Cell Preparation

Centrifuge cells at 300 g / 5 min, wash with Opti-MEM, and resuspend serum-free at the recommended density for your format.

3

Transfection

Mix complex with cell suspension. Incubate 15–30 minutes in a standard cell culture incubator — no electroporator required.

4

Culture & Assess

Add complete medium and culture. Assess editing efficiency by flow cytometry or TIDE at 48–72 hours.
