1 vial of 100 µg whole HEK293T cell lysate in RIPA buffer
Storage The lysate is shipped with dry ice. Upon receiving, store the sample at -80°C. Also after dilution, the protein sample should be aliquoted and stored at -80°C for long term storage. Avoid repeated freeze-thaw cycles. Lysate samples can be diluted with 1x SDS-PAGE running buffer (with reducing agent). Lysate samples are stable for 12 months from the date of receipt when stored at -80°C. Lysate samples are stable for 12 months from the date of receipt when stored at -80°C.

Preparation HEK293T cells in 10-cm dishes were transiently transfected with MegaTran Transfection Reagent (TT200002) and 5ug TrueORF cDNA plasmid. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix (Sigma), 1mM PMSF and 1mM Na3VO4), and then centrifuged to clarify the lysate. Protein concentration was measured by BCA kit (Thermo Scientific Inc.).

AI Summary: OriGene contributed to this study by providing the pLenti-C-Myc-DDK lentiviral gene expression vector (PS100064), GAPDH Myc-DDK lentivirus plasmid (RC202309L1), and PGK1 Myc-DDK lentivirus plasmid (RC211172L1) used for overexpression experiments to investigate glycolytic bottlenecks in stem cell-derived β cells.
Snippet: … Lentiviral Overexpression of GAPDH and PGK1 All overexpression plasmids were purchased from Origene as frozen plasmids, all derived from the parental pLenti-C-Myc-DDK backbone (RC202309L1). 293T cells were plated in Poly-L-Lysine-coated 15 cm2 dishes and lipofected using Trans-IT 293 reagent;aqueous fraction was dried and resuspended in 1% acetic acid in water, pH 9 to preserve NAD redox state. AI Summary: OriGene’s full-length human PGK1 (NM_000291) plasmid was used for transient expression of PGK1 protein, enabling functional studies that linked glycolysis to KEAP1-NRF2 signalling in this research.
Snippet: Full-length, human PGK1 (NM_000291, Origene) transiently expressed from a pCMV6 entry vector with a C-terminal Myc-DDK tag;full-length, human KEAP1 (28023, Addgene) was transiently expressed from a pcDNA/FRT/TO plasmid with a C-terminal 3×Flag tag. All references to Flag–PGK1 or Flag–KEAP1 represent the proteins in the aforementioned vectors, respectively
