1 vial of 100 µg whole HEK293T cell lysate in RIPA buffer
Storage The lysate is shipped with dry ice. Upon receiving, store the sample at -80°C. Also after dilution, the protein sample should be aliquoted and stored at -80°C for long term storage. Avoid repeated freeze-thaw cycles. Lysate samples can be diluted with 1x SDS-PAGE running buffer (with reducing agent). Lysate samples are stable for 12 months from the date of receipt when stored at -80°C. Lysate samples are stable for 12 months from the date of receipt when stored at -80°C.

Preparation HEK293T cells in 10-cm dishes were transiently transfected with MegaTran Transfection Reagent (TT200002) and 5ug TrueORF cDNA plasmid. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix (Sigma), 1mM PMSF and 1mM Na3VO4), and then centrifuged to clarify the lysate. Protein concentration was measured by BCA kit (Thermo Scientific Inc.).

Dihydrotestosterone inhibits arylsulfatase B and Dickkopf Wnt signaling pathway inhibitor (DKK)-3 leading to enhanced Wnt signaling in prostate epithelium in response to stromal Wnt3A
Bhattacharyya, Feferman, Tobacman
Prostate (2019)
AI Summary: OriGene’s ARSB (TrueClone, NCBI NM_000046, pCMV6-XL4 vector) plasmid was used to overexpress ARSB in prostate epithelial cells, enabling the study of ARSB’s role in regulating Wnt signaling and DKK3 expression in the context of prostate cancer.
Snippet: Skip to Main Content;Overexpression of ARSB (NCBI NM_000046, transcript variant 1;TrueClone, OriGene, Rockville, MD) was performed by transfection of specific untagged plasmids in pCMV6‐XL4 vector using 2 µg of the plasmid and Lipofectamine 2000 (Invitrogen, Carlsbad, CA). …
Chondroitin sulfatases differentially regulate Wnt signaling in prostate stem cells through effects on SHP2, phospho-ERK1/2, and Dickkopf Wnt signaling pathway inhibitor (DKK3)
Bhattacharyya, Feferman, Tobacman
Oncotarget (2017) 8 (59), 100242-100260 DOI: 10.18632/oncotarget.22152
Snippet: Cells were silenced for either ARSB (EC 3.1.6.12) or GALNS (EC 3.1.6.4) by specific siRNA (Qiagen, Germantown, MD) (see below). Overexpression of ARSB (NCBI NM_000046, transcript variant 1;TrueClone, OriGene, Rockville, MD) or of GALNS (NCBI NM_000512;TrueClone, Origene) was performed by transfection of specific untagged plasmids (see below), as previously [57]
