OTI Disclaimer The molecular sequence of this clone aligns with the gene accession number as a point of reference only. However, individual transcript sequences of the same gene can differ through naturally occurring variations (e.g. polymorphisms), each with its own valid existence. This clone is substantially in agreement with the reference, but a complete review of all prevailing variants is recommended prior to use. More info
OTI Annotation This clone was engineered to express the complete ORF with an expression tag. Expression varies depending on the nature of the gene.
Components The ORF clone is ion-exchange column purified and shipped in a 2D barcoded Matrix tube containing 10ug of transfection-ready, dried plasmid DNA (reconstitute with 100 ul of water).
Reconstitution Method 1. Centrifuge at 5,000xg for 5min.
2. Carefully open the tube and add 100ul of sterile water to dissolve the DNA.
3. Close the tube and incubate for 10 minutes at room temperature.
4. Briefly vortex the tube and then do a quick spin (less than 5000xg) to concentrate the liquid at the bottom.
5. Store the suspended plasmid at -20°C. The DNA is stable for at least one year from date of shipping when stored at -20°C.
Note Plasmids are not sterile.  For experiments where strict sterility is required, filtration with 0.22um filter is required.

Non-industrial production of therapeutic lentiviral vectors: How to provide vectors to academic CAR-T
Keyer, Syzdykova, Ingirbay et al
Biotechnol Bioeng (2024)
Cited by 6
AI Summary: OriGene’s pLenti-C-Myc-DDK-IRES-Puro plasmid (Cat. PS100069) was used as the parental lentiviral vector backbone in this study to optimize and scale up the academic production of therapeutic lentiviral vectors for CAR-T cell generation.
Snippet: Parent LV is from pLenti-C-Myc-DDK-IRES-Puro (OriGene Cat. PS100069). The parental plasmid already contains a self-inactivating LV with a viral 5′-LTR (R-U5) downstream of the CMV promoter, and 3′-LTR with a deletion (deltaU3-R-U5)
TL1A is an epithelial alarmin that cooperates with IL-33 for initiation of allergic airway inflammation

AI Summary: In this study, the OriGene mTL1A-Flag expression vector (catalog #MR203224) was used to ectopically express mouse TL1A protein in epithelial cells, enabling the authors to investigate TL1A's role as an epithelial alarmin in allergic airway inflammation.
Snippet: tor allowing ectopic expression of the mouse TL1A protein (252 aa) tagged at its C-terminus with the epitope Myc-DDK (Flag), under the control of a CMV promoter (mTL1A-Flag vector), was obtained from Origene Technologies (# MR203224). Plasmid pcDNA3.1 (CMV-promoter) was used as a control vector. The U2OS epithelial cell line (# HTB-96;ATCC), an adherent epithelial cell line with 20–60% transfec
