Summary IFIH1 encodes MDA5 which is an intracellular sensor of viral RNA that triggers the innate immune response. Sensing RNA length and secondary structure, MDA5 binds dsRNA oligonucleotides with a modified DExD/H-box helicase core and a C-terminal domain, thus leading to a proinflammatory response that includes interferons. It has been shown that Coronaviruses (CoVs) as well as various other virus families, are capable of evading the MDA5-dependent interferon response, thus impeding the activation of the innate immune response to infection. MDA5 has also been shown to play an important role in enhancing natural killer cell function in malaria infection. In addition to its protective role in antiviral responses, MDA5 has been implicated in autoimmune and autoinflammatory diseases such as type 1 diabetes, systemic lupus erythematosus, and Aicardi-Goutieres syndromeprovided by RefSeq, Jul 2020

A non-radioactive modified immunoprecipitation assay for diagnostic detection of anti-MDA5 antibodies in idiopathic inflammatory myopathies
Wang, Teo, Lan et al
Front Immunol (2026) 17, 1896104-1896104 DOI: 10.3389/fimmu.2026.1896104
AI Summary: In this study, OriGene's recombinant human MDA5 protein (cat. no. TP315661) was used as a positive protein control to validate the modified immunoprecipitation assay for detecting anti-MDA5 antibodies in idiopathic inflammatory myopathies.
Snippet: g at approximately 95 °C for 10 min.Immunoprecipitated proteins were separated by Bis-Tris SDS-PAGE and transferred onto polyvinylidene difluoride membranes. Recombinant human MDA5 protein (OriGene Technologies;cat. no. TP315661) was used as a positive protein control. Immunoprecipitated MDA5 was detected using a rabbit monoclonal anti-MDA5 antibody (clone D74E4;Cell Signaling Technolo
Increased Serum Matrix Metalloproteinase-9 Levels are Associated with Anti-Jo1 but not Anti-MDA5 in Myositis Patients
Liu, Luo, Wang et al
Aging Dis (2019) 10 (4), 746-755 DOI: 10.14336/AD.2018.1120
AI Summary: OriGene’s recombinant full-length MDA5 antigen was used to coat ELISA plates for the detection and quantification of anti-MDA5 antibodies in patient serum, enabling the study’s analysis of serological differences in myositis patients.
Snippet: Briefly, 96-well microplates were coated with 0.5 µg/ml recombinant full-length MDA5 antigen (Origene Technologies, MD, USA) overnight at 4°C. …The antibody units were calculated from the optimal densities at 450 nm using a standard curve obtained from serial concentrations of serum containing anti-MDA5 antibodies
