• Background: Efficient Western blotting starts with high-quality protein extraction. Standard methods rely on detergent-based buffers like RIPA (0.1% SDS) or NP-40 (SDS-free) and physical disruption via sonication. While RIPA buffer effectively lyses mammalian cells and tissues, it struggles to extract large proteins, often requiring sonication to improve yield. However, excessive sonication can degrade large proteins, and additional inhibitors are needed to prevent enzymatic degradation. IntactProtein Cell-Tissue Lysis Buffer streamlines this process by eliminating the need for separate protease and phosphatase inhibitors while preserving post-translational modifications (PTMs). Designed for optimal recovery of proteins of all sizes, it ensures reliable extraction from mammalian cells and tissues, enhancing Western blot performance.
• Components: 1 vial of 40 uL Reagent A 1 vial of 20 mL Reagent B
• Stability: 1. Reagent B can be stored at room temperature for up to 3 months. For long-term storage, keep the Reagent B at 4 °C. Do not freeze. 2. Precipitation may occur when Reagent B is stored at 4 °C over a prolonged period of time. Precipitation does not affect product quality. Precipitation will redissolve at room temperature; after the precipitation disappears, gently mix the solution.
