Product Data
Applications ELISA, IP, WB
Recommended Dilution Suitable for Immunoblotting (Western or Dot blot), ELISA, Immunoprecipitation, conjugation and most immunological methods requiring high titer and specificity.
Recommended Dilutions:
ELISA: 1/5,000-1/25,000.
Western Blot: 1/500-1/3,000.
Note: This product has been assayed by immunoblot against CIITA using Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] (Goat) and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:1,000 is suggested for this product.
Best Results are obtained using 0.03% (v/v) Triton X-100 IN BUFFERS.
Do not supplement buffers with Tween-20 detergent.
Reactivities Human
Host Rabbit
Clonality Polyclonal
Immunogen Peptide corresponding to a region near the C-terminal end (amino acids 726-742) of the human CIITA gene conjugated to Keyhole Limpet Hemocyanin (KLH).
Specificity This product is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above.
Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum.
Cross reactivity against CIITA from other sources may occur but has not been specifically determined.
Formulation 0.02M Potassium Phosphate, 0.15M Sodium Chloride, pH 7.2 with 0.01% Sodium Azide as preservative.
State: Ig Fraction
State: Liquid (sterile filtered) purified IgG fraction.
Concentration lot specific
Purification Multi-step process.
Conjugation Unconjugated
Storage Store the antibody (undiluted) at 2-8°C for one month or (in aliquots) at -20°C for longer.
Dilute only prior to immediate use.
Avoid cycles of freezing and thawing.
Stability Shelf life: One year from despatch.
Gene Name class II, major histocompatibility complex, transactivator
Background The transactivator CIITA regulates basal and interferon-induced expression of MHC class II genes. CIITA restores expression of all MHC II gene expression in mutant cells and corrects regulatory defects of those genes. Activation is via the proximal promoter. No DNA binding of in vitro translated CIITA was detected. CIITA may act in a coactivator-like fashion through protein-protein interactions by contacting factors binding to the proximal MHC class II promoter, to elements of the transcription machinery, or both. Alternatively it may activate HLA class II transcription by modifying proteins that bind to the MHC class II promoter.
Synonyms MHC2TA
