Summary This gene belongs to the forkhead family of transcription factors which are characterized by a distinct forkhead domain. This gene likely functions as a trigger for apoptosis through expression of genes necessary for cell death. Translocation of this gene with the MLL gene is associated with secondary acute leukemia. Alternatively spliced transcript variants encoding the same protein have been observed. provided by RefSeq, Jul 2008
shRNA Design These shRNA constructs were designed against multiple splice variants at this gene locus. To be certain that your variant of interest is targeted, please contact techsupport@origene.com. If you need a special design or shRNA sequence, please utilize our custom shRNA service.

β-amyloid-induced gonadotropin-releasing hormone decline involving Forkhead transcription factor FOXO3a and nuclear factor-κB
Shi, Shi, Guo et al
Neuroreport (2020) 31 (12), 923-927 DOI: 10.1097/WNR.0000000000001488
AI Summary: OriGene’s FOXO3a shRNA-expressing plasmid was used in this study to specifically silence FOXO3a and investigate its role in β-amyloid-induced gonadotropin-releasing hormone decline.
Snippet: The FOXO3a shRNA-expressing plasmid was obtained from Origene Technologies, Inc (Rockville, Maryland, USA). The ‘scrambled’ control, the packaging plasmid (pCMV-dr8.2 dvpr) and the envelop plasmid (pCMV-VSV-G) were from Miaoling Biotechnology, Inc (Wuhan, HuBei, China). Lentivirus-mediated FOXO3a-shRNA silencing was performed according to our published procedure [8,9]
Reprogramming ovarian and breast cancer cells into non-cancerous cells by low-dose metformin or SN-38 through FOXO3 activation
Hu, Chung, Guan et al
Sci Rep (2014) 4, 5810 DOI: 10.1038/srep05810

AI Summary: In this study, OriGene’s FOXO3 shRNA lentiviral particles and FOXO3 overexpression plasmids were used to manipulate FOXO3 expression, enabling the authors to demonstrate that FOXO3 activation is essential for reprogramming ovarian and breast cancer cells into non-cancerous cells by low-dose metformin or SN-38.
Snippet: FOXO3 is a key transcription factor that controls the transcription of a number of genes crucial for regulating cell cycle control18, DNA damage and stress responses19, 20, 21, 22, aging and longevity23, 24, cellular apoptosis25, 26, 27, and suppression of cancer28, 29, 30, 31 in animal and human cells. … While multiple mechanisms have been shown to regulate FOXO3 activity, phosphorylation inhibits FOXO3 nuclear translocation that is central for its regulation and function. This nuclear exclusion and translocation of FOXO3 into the cytoplasm inhibits FOXO3-dependent transcription. Loss of function of FOXO3 has been linked to tumorigenesis and poor patient survival in BCa28, 30, 31, and OvCa34 suggesting that FOXO3 is a pivotal tumor suppressor

AMPK Reverses the Mesenchymal Phenotype of Cancer Cells by Targeting the Akt-MDM2-Foxo3a Signaling Axis
Chou, Lee, Lai et al
Cancer Res. (2014)
AI Summary: In this study, OriGene's Foxo3a shRNA was used to silence Foxo3a expression, enabling the authors to investigate the role of Foxo3a in mediating the effects of AMPK on the mesenchymal phenotype of cancer cells.
Snippet: The plasmids encoding constitutively active (CA)-Akt, pcDNA-MDM2, and HA-ubiquitin were obtained from Addgene (Cambridge, MA), and Foxo3a shRNA from Origene (Rockville, MD). Foxo3a-GFP plasmid was kindly provided by Dr. Mickey CT. ... Cite SaveSaving
