Summary The protein encoded by this gene is a regulator of apoptosis and is structurally similar to caspase-8. However, the encoded protein lacks caspase activity and appears to be itself cleaved into two peptides by caspase-8. Several transcript variants encoding different isoforms have been found for this gene, and partial evidence for several more variants exists. provided by RefSeq, Feb 2011
shRNA Design These shRNA constructs were designed against multiple splice variants at this gene locus. To be certain that your variant of interest is targeted, please contact techsupport@origene.com. If you need a special design or shRNA sequence, please utilize our custom shRNA service.

Esophageal cancer cells convert the death signal from TRAIL into a stimulus for survival during acid/bile exposure
Chang, Dang, Che et al
Dig Liver Dis (2020) 52 (10), 1195-1200 DOI: 10.1016/j.dld.2020.04.013

AI Summary: In this study, OriGene products—including TRADD (Cat# RC213682) and c-FLIP (Cat# RC211744) expression plasmids—were used to investigate how esophageal cancer cells resist TRAIL-induced cell death by overexpressing these proteins and altering apoptotic signaling pathways.
TNF Apoptosis Protection Fraction (TAPF) prevents apoptosis induced by TNF, but not by Fas or TRAIL, via NF-κB-induced increase in cFLIP
Seol, Mihich, Berleth
Cytokine (2015)
Cited by 12
AI Summary: In this study, OriGene's plasmids encoding 29-mer shRNAs targeting cFLIP and a non-specific shRNA (targeting GFP) were used to decrease cFLIP levels in human dermal fibroblasts, helping to demonstrate that TAPF's protection against TNF-induced apoptosis depends on cFLIP expression.
Snippet: 2.8. Retroviral Infection of NHDF with shRNA. Plasmids encoding three different 29 mer short hairpin RNAs (shRNAs) targeting cFLIP, one encoding a non-specific shRNA (targeting GFP) and a vector control were purchased (Origene, Rockville MD). ... Cite SaveSaving
Inhibition of NF-kappaB signaling by quinacrine is cytotoxic to human colon carcinoma cell lines and is synergistic in combination with tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) or oxaliplatin
Jani, DeVecchio, Mazumdar et al
J Biol Chem (2010) 285 (25), 19162-72 DOI: 10.1074/jbc.M109.091645
AI Summary: OriGene's shRNA constructs targeting c-FLIP and Mcl-1 were used to knock down these genes in colon carcinoma cell lines, demonstrating that reduced expression of these NF-kappaB-regulated survival factors significantly increased sensitivity to TRAIL and oxaliplatin-induced cell death.
