Summary This gene encodes a protein that is part of a post-splicing multiprotein complex involved in both mRNA nuclear export and mRNA surveillance. mRNA surveillance detects exported mRNAs with truncated open reading frames and initiates nonsense-mediated mRNA decay (NMD). When translation ends upstream from the last exon-exon junction, this triggers NMD to degrade mRNAs containing premature stop codons. This protein is located only in the cytoplasm. When translation ends, it interacts with the protein that is a functional homolog of yeast Upf2p to trigger mRNA decapping. Use of multiple polyadenylation sites has been noted for this gene. Alternative splicing results in multiple transcript variants. provided by RefSeq, Jul 2014
shRNA Design These shRNA constructs were designed against multiple splice variants at this gene locus. To be certain that your variant of interest is targeted, please contact techsupport@origene.com. If you need a special design or shRNA sequence, please utilize our custom shRNA service.

UPF1 silenced cellular model systems for screening of read-through agents active on β039 thalassemia point mutation
Salvatori, Pappadà, Breveglieri et al
BMC Biotechnol. (2018) 18 (1), 28 DOI: 10.1186/s12896-018-0435-0
AI Summary: OriGene products pRS-shRNA-UPF1 and pRS-shRNA-scramble retroviral vectors were used to obtain shRNA and U6 promoter sequences for generating UPF1-silenced cellular models in this study.
Snippet: of the U6 promoter. shRNA and U6 promoter sequences were obtained from commercial retroviral vectors: pRS-shRNA-UPF1 and pRS-shRNA-scramble (Origene Technologies, Rockville, MD, USA). The lentiviral constructs Cached

AI Summary: OriGene products, specifically the UPF1 shRNA pool (TR308482) and GFP-specific control shRNA plasmid, were used to modulate UPF1 expression in HeLa cells, enabling the authors to assess the role of UPF1 in nonsense-mediated decay within their luciferase reporter assay.
Snippet: 2.8. UPF1 knockdown and overexpression. To knock down the expression of UPF1, HeLa cells were transiently transfected with a pool of four UPF1 shRNAs (Origene;TR308482) or a GFP-specific control shRNA plasmid (Origene).
