Background
CD33 or Siglec-3 (sialic acid binding Ig-like lectin 3) is a transmembrane receptor expressed on cells of myeloid lineage. It binds sialic acids and is a member of the SIGLEC family of lectins. CD33 expression is restricted to early multilineage hematopoietic progenitors, myelomonocytic precursors and more mature myeloid cells, being absent on normal pluripotent hematopoietic stem cells. CD33 can be stimulated by any molecule with sialic acid residues such as glycoproteins or glycolipids, which results in a cascade that inhibits phagocytosis in the cell. About 85–90% of acute myeloid leukemia cases express the CD33 antigen. Circulating CD33 (cCD33) can be detected in the plasma from patients with leukemias, and cCD33 levels may have clinical implication such as predictive and prognostic value in these patients.
