Clone Name AT4D3
Application ELISA, FC, ICC, IHC, WB
Recommended Dilution ELISA.
Western blot (1/1,000-1/2,000).
Cell lysates of HeLa (40 µg) were resolved by SDS-PAGE, transferred to NC membrane and probed with anti-human FkBP4 (1/1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
Immunohistochemistry on Paraffin Embedded Tissue (1/50-1/100).
Paraffin embedded sections of Seminoma (Germ cell tumor) tissue were incubated with anti-Human FKBP4 (1/50) for 2 hours at RT. Antigen retrieval was performed in 0.1M sodium citrate buffer and detected using Diaminobenzidine (DAB).

Background FkBP4, also known as FkBP52, is a member of the immunophilin protein family, which play a role in immunoregulation and basic cellular processes involving protein folding and trafficking. It is a component of unactivated mammalian steroid receptor complexes and may play a role in the intracellular trafficking of hetero-oligomeric forms of the steroid hormone receptors. It interacts with interferon regulatory factor-4 and plays an important role in immunoregulatory gene expression in B and T lymphocytes.

AI Summary: In this study, OriGene’s FKBP52 shRNA (TG312896) and FKBP52 cDNA (RC217709) products were used to manipulate FKBP52 expression in glioblastoma cells, enabling the authors to demonstrate FKBP52’s key role in regulating TDO-mediated tryptophan catabolism.
