Immunogen | Immune sera was collected from goats that were hyper-immunized with purified chicken IgY. Production: Goat anti-Chicken IgY antibodies were affinity purified from the sera over a Chicken IgY column, and then conjugated to N-hydroxysuccinimide (NHS)-activated Agarose beads. After blocking residual NHS sites, the agarose beads were washed exhaustively with 50mM HEPES buffer (pH 7.6) with 5mM EDTA (TE buffer). Sodium Azide was then added to a final concentration of 0.02% (w/v). |
Specificity | Binding Capacity: 1.0 mg of Chicken IgY per ml bed volume. Quality Control: The capacity of PrecipHen® was determined by measuring the 2.5 mls of PrecipHen. One milliliter aliquots of chicken IgY (200 µg/ml in TE buffer) were applied to the column and fractions were collected. A similar column of agarose (non-NHS activated) was used for comprison. The concentration of IgY passing through each column was measured, and the total amount of IgY that bound to the column was calculated. |
