HEK293T cells were transfected with the pCMV6-ENTRY control (Cat# PS100001, Left lane) or pCMV6-ENTRY BACH1 (Cat# RC221628, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-BACH1(Cat# TA504484). Positive lysates LY404144 (100ug) and LC404144 (20ug) can be purchased separately from OriGene.

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type HeLa cells (WT, Cat# LC810HELA) and BACH1-Knockout HeLa cells (KO, Cat# LC835403) were separated by SDS-PAGE and immunoblotted with anti-BACH1 monoclonal antibody TA504484 (1:2000). Then the blotted membrane was stripped and reprobed with anti-HSP90 antibody as a loading control.

  HEK293T cells transfected with either RC221628 overexpress plasmid (Red) or empty vector control plasmid (Blue) were immunostained by anti-BACH1 antibody (TA504484), and then analyzed by flow cytometry.

• Background: This gene encodes a transcription factor that belongs to the cap'n'collar type of basic region leucine zipper factor family (CNC-bZip). The encoded protein contains broad complex, tramtrack, bric-a-brac/poxvirus and zinc finger (BTB/POZ) domains, which is atypical of CNC-bZip family members. These BTB/POZ domains facilitate protein-protein interactions and formation of homo- and/or hetero-oligomers. When this encoded protein forms a heterodimer with MafK, it functions as a repressor of Maf recognition element (MARE) and transcription is repressed.
