Gene Name phosphatase and tensin homolog
Background Thephosphatase and tensin homologue (PTEN) tumor suppressoris a phosphatidylionositol D3-phosphatase that contains the catalytic signature motif (HCXXGXXRS/T) found in all members of the protein tyrosine phosphatase family. PTEN is mutated in a wide range of cancers.In vitro, the recombinant PTEN can be regulated by phosphorylation on multiple serine and threonine residues in its C-terminus, such as CK2, GSK3beta at Ser-362, 370 and 385. It antagonizes signal transduction downstream of PI-3 kinases affecting cell proliferation, apoptosis and migration. Loss-of-function mutations in the PTEN gene lead to constitutive activation of multiple signaling pathways.
