Recommended Dilution Direct ELISA: To detect Mouse IL-10 by Direct ELISA (using 100 μl/well antibody solution) a concentration of approx. 1.0 μg/ml of this antibody is required. This Biotin conjugated antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4 ng/well of recombinant Mouse IL-10.
Sandwich ELISA: To detect Mouse IL-10 by Sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.25-1.0 μg/ml of this antibody is required. This Biotin conjugated antibody, in conjunction with Purified Anti-Murine IL-10 antibody (Cat.-No PP007P1 or PP007P2) as a Capture antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Mouse IL-10.
Western Blot: To detect Mouse IL-10 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Mouse IL-10 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Gene Name interleukin 10
Background Interleukin 10 (IL-10) is a 178 amino acid protein that is primarily secreted by TH2 clones. IL-10 has dual functions, the first of which is the suppression of cytokine production by TH1 clones responding to antigen presented by monocyte and macrophage antigen presenting cells (APCs). The second function consists of the inhibition of response of cytokine targeted cells, possibly by the downregulation of CD25 (the interleukin-2 receptor) on macrophages and B lymphocytes. IL-10 exerts its biological activity through the IL-10 receptor (IL-10R); a 560 amino acid, 110kDa glycoprotein whose expression can be induced in cultured macrophages and fibroblasts by elevated in HIV-1 infected individuals and has been implicated in the progression of the disease.
