HEK293T cells were transfected with the pCMV6-ENTRY control (Cat# PS100001, Left lane) or pCMV6-ENTRY TARBP2 (Cat# RC201043, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-TARBP2 (Cat# TA808481)(1:2000). Positive lysates LY418167 (100ug) and LC418167 (20ug) can be purchased separately from OriGene.

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type HeLa cells (WT, Cat# LC810HELA) and TARBP2-Knockout HeLa cells (KO, Cat# LC833009) were separated by SDS-PAGE and immunoblotted with anti-TARBP2 monoclonal antibody TA808481 (1:500`). Then the blotted membrane was stripped and reprobed with anti-HSP90 antibody as a loading control.

• Background: HIV-1, the causative agent of acquired immunodeficiency syndrome (AIDS), contains an RNA genome that produces a chromosomally integrated DNA during the replicative cycle. Activation of HIV-1 gene expression by the transactivator Tat is dependent on an RNA regulatory element (TAR) located downstream of the transcription initiation site. The protein encoded by this gene binds between the bulge and the loop of the HIV-1 TAR RNA regulatory element and activates HIV-1 gene expression in synergy with the viral Tat protein.
