Background | Caspase-9 belongs to a member of the cysteine-aspartic acid protease (Caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small. Those dimerize to form the active enzyme. Activation of procaspase-9 by Apaf-1 in the Cytochrome C/dATP-dependent pathway requires proteolytic cleavage to generate the mature caspase molecule. Once activated, caspase-9 can initiate a caspase cascade involving the downstream executioner caspase-3, -6 and -7. |
