Recommended Dilution Can be used in precipitating techniques as Immunoelectrophoresis and Radial Immunodiffusion to identify the presence of IgM in duck serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of Duck IgM from serum or plasma.
Recommended Dilutions:
Immunoelectrophoresis: Use 2 µl or equivalent against 120 µl antiserum.
Double Radial Immunodiffusion (Ouchterlony): Use a rosette arrangement with 10 µl in a 3 mm diameter center well and 2 µl serum samples (neat and serially diluted) in 2 mm diameter peripheral wells.
Precipitin titre: 1/16 when tested against pooled normal duck serum in agar-block immunodiffusion titration.

Freund's complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is restricted to the Fc part of the IgM molecule. In Immunoelectrophoresis and Radial Immunodiffusion, using various antiserum concentrations against normal Duck serum a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgM. No precipitation reaction is obtained with purified IgG, IgA, and IgG/Fab fragments.
The antiserum does not cross-react with any other component of the Duck Ig system. Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.

Reconstitution Method Restore by adding 1 ml of sterile distilled water.
Concentration Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.

Storage Store lyophilized at 2-8°C and reconstituted at 2-8°C for one month or (in aliquots) at -20°C for longer.
Avoid Repeated thawing and freezing.
Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required to eliminate antibodies crossreacting with other components of the immunoglobulin system or reacting with other serum proteins. Special attention is given to the removal of antibodies to common Ig/Fab. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
