Gene Name death associated protein 3
Background Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Recently, five novel apoptotic genes (named DAP genes for: Death Associated Proteins) have been identified that code for proteins, which display a diverse spectrum of biochemical activities. These include a novel type of calcium/calmodulin-regulated kinase which carries ankyrin repeats and a death domain (DAP-kinase), a nucleotide-binding protein (DAP-3), a small proline-rich cytoplasmic protein (DAP-1), and a novel homolog of the eIF4G translation initiation factor (DAP-5). DAP-3 has an ATP/GTP binding motive (P-loop) and has been isolated from human and mouse, and named as hDAP-3 and mDAP-3 (Death Associated Protein-3), respectively. Sequence analysis revealed an 82% amino acid identity between human and mouse protein. In situ hybridization and northern blot analysis showed an abundant mRNA expression with a pronounced expression in highly proliferative epithelial compartments. Overexpression of DAP-3 leads to cytochrome c release and induction of cell death. DAP-3 mediated apoptosis was shown to depend on a functional P-loop. mDAP-3 is localized to the mitochondrial matrix and in contrast to cytochrome c, retained its mitochondrial localization during apoptosis induction.
