Sandwich ELISA: To detect Human IL-33 by Sandwich ELISA (using 100μl/well antibody solution) a concentration of 0.5-2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human IL-33 antibody (Cat.-No AP32086BT-S or AP32086BT-N) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Human IL-33.
Western Blot: To detect Human use this antibody at a concentration of 0.1-0.2 μg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human IL-33 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
Neutralization: 
To yield one-half maximal inhibition [ND50] of the biological activity of Human IL-33 (1.0 ng/ml), a concentration of 0.2-0.4 μg/ml of this antibody is required.
Immunohistochemistry on Paraffin Sections: 
0.125-0.25 μg/ml with overnight incubation at 4°C. Staining of Formalin-Fixed, Paraffin Embedded Sections of Human breast malignant ductal adenocarcinoma using an HRP labeled polymer detection system with a DAB Chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended. Tissue samples were provided by the Cooperative Human Tissue Network, which is funded by the National Cancer Institute.

Gene Name interleukin 33
Background Interleukin 33 (IL-33) is a 32 kDa proinflammatory cytokine and intracellular nuclear factor with transcriptional regulatory properties. IL-33 is structurally related to IL-1, which induces helper T-cells to produce type 2 cytokines and acts through the receptor IL1RL-1 (IL1 receptor-like-1), which is known also as ST2. Binding of IL-33 to this receptor activates NF-kappa-B and MAP kinases and induces in vitro Th2 cells to produce cytokines. In vivo, IL-33 induces expression of IL-4, IL-5, IL-13 and leads to severe pathological changes in mucosal organs and in vitro, it can be divided to N-terminal fragment of 12 kDa and C-terminal fragment of 18kDa by cleavage of caspase-1.
