Recommended Dilution The fluorescent immunoconjugate to Human C5 is used to determine the presence and pattern using Immunohistochemical staining techniques. Determinations of individual C components can be very useful in defining the exact location of a defect.
This immunoconjugate is not pre-diluted. The optimum working dilution of each conjugate should be established by titration before being used. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal.
Working Dilutions: 1/10-1/40.

Specificity The reactivity of the antiserum is restricted to C5.
In immunoelectrophoresis and Radial Immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified C5 protein. No reaction is obtained with any other plasma protein component or serum.
The antiserum does not cross react with any other component of human plasma. Inter-species crossreactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.

Storage Store lyophilized at 2-8°C for 6 months or at -20°C long term.
After reconstitution store the antibody undiluted at 2-8°C for one week or (in aliquots) at -20°C long term. Avoid repeated freezing and thawing.
This product is photosensitive and should be protected from light.

Gene Name complement component 5
Background C5 component of human complement is a plasma protein consisting of two polypeptide chains, alpha, beta, bound by disulphide bonds. It has a molecular weight of 190,000. The activated protein forms with the complement factors C6-C9 the membrane attack complex.

Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other dog serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
