• Recommended Dilution: Immunoprecipitation.
  Can be used in precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion to identify the presence hemopexin in human serum or other body fluids and to determine its concentration. This antiserum has not been tested in more sensitive assays as nephelometry, ELISA or use in immunohistology, but this does not exclude such use if proper controls are included.
  Directions for use: In immunoelectrophoresis in agarose-plates use 2 µl human plasma or equivalent against 120 µl antiserum. In double radial immunodiffusion use a rosette arrangement with 10 µl antiserum in 3 mm diameter center well and 2 µl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells.
  Precipitin Titre: 1/32 when tested against pooled normal human serum in agar-block immunodiffusion titration.

• Specificity: The reactivity of the antiserum is restricted to hemopexin as tested in immunoelectrophoresis and radial immunodiffusion. A characteristic single precipitin line is obtained with normal serum which shows a reaction of identity with the precipitin line with purified hemopexin.
  Cross-reactivity: Inter-species cross-reactivity is a normal feature of antibodies to serum proteins, since homologous proteins of different species frequently share antigenic determinants. Cross reactivity of this antiserum has not been tested in detail. In double radial immunodiffusion no reaction has been observed with rat and mouse.

• Background: Hemopexin is a haem-binding b-globulin present in normal serum at a concentration of 0.8 to 1.0 mg/ml. Its molecular weight is 57,000. The carbohydrate content is 23%. It consists of one polypeptide chain.
