SET7/ SET9 is a histone methyltransferase that can monomethylate Lysine 4 of histone H3 and regulate protein expression (1, 2). This methylation represents a specific tag for epigenetic transcriptional activation. SET7/ SET9 plays a central role in the activation of proteins such as collagenase or insulin. It is recruited by IPF1/PDX-1 to the insulin promoter, activating transcription. Moreover, SET7/ SET9 has methyltransferase activity toward non-histone proteins such as p53/TP53, TAF10, and possibly TAF7 by recognizing and binding the [KR]-[STA]-K in substrate proteins (2).
