FunctionABCA1 is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intracellular membranes, and are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the ABC1 subfamily. Members of the ABC1 subfamily comprise the only major ABC subfamily found exclusively in multicellular eukaryotes. With cholesterol as its substrate, ABCA1 functions as a cholesterol efflux pump in the cellular lipid removal pathway. Mutations in this protein have been associated with Tangier's disease and familial high-density lipoprotein deficiency (1). It has been shown that Ser-1042 and Ser-2054, located in the nucleotide binding domains of ABCA1, are major phosphorylation sites for PKA. Phosphorylation of Ser-2054 within the nucleotide binding domain 2 was found to be particularly important for ApoA-I-dependent phospholipid efflux activity mediated by ABCA1 (2).
