FunctionHomeodomain-interacting protein kinase 2 (HIPK2) is a conserved serine/threonine nuclear kinase that interacts with homeodomain transcription factors and acts as a corepressor of several transcription factors, including SMAD1 and POU4F1/Brn3a and probably NK homeodomain transcription factors (1). In most cell types, HIPK2 activation is associated with the induction of growth suppression and apoptosis, which is mediated through both p53-dependent and p53-independent HIPK2 signaling routes. HIPK2 was found to be a DNA damage-responsive kinase that gets activated upon UV, ionizing irradiation and chemotherapeutic drug treatment (2). In response to severe DNA damage, HIPK2 forms a complex with p53 and phosphorylates p53 at Ser46, which in turn activates apoptotic p53-target genes such as PUMA, Bax, and Noxa (3). Upon DNA damage in the absence of p53, HIPK2 phosphorylates anti-apoptotic corepressor C-terminal binding protein (CtBP) at Ser422, leading to proteasomal degradation of CtBP (4). HIPK2 activity can be restrained by relocalizing into the cytoplasm mediated by HMGA1 (5) or the E3 ubiquitin ligases Siah-1 and WSB-1-mediated proteasome-dependent degradation (6,7).
