Cyclin-dependent kinase 2-associated protein 1 (CDK2AP1), also known as DOC-1, is a tumor suppressor in the CDK2AP family (1). CDK2AP1 is expressed in all normal human tissues, and it is an inhibitor of G(1)/S transition through the down-regulation of CDK2 (1, 2). As an essential gene for early embryonic development, CDK2AP1 also associates with DNA polymerase α/primase and mediates the phosphorylation of the large p180 catalytic subunit, suggesting that it may be a potential regulator of DNA replication in the S phase of the cell cycle (1, 2). It is a core subunit of MBD2/NuRD and MBD3/NuRD, which is involved in DNA methylation (3).
