FunctionProlyl hydroxylase domain-containing protein 1 (PHD1, Hypoxia-inducible factor prolyl hydroxylase 1) is a 2-oxoglutarate and dioxygen-dependent enzyme that mediates the rapid destruction of hypoxia-inducible factor (HIF) alpha subunits by catalyzing their post-translational formations (1). PHD1 is in the Egln family of proline hydroxylases, functioning as a cellular oxygen sensor (2). Under normoxic conditions, it targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitination complex (2). Loss of PHD1 has been shown to lower oxygen consumption in skeletal muscle by reprogramming glucose metabolism from oxidative to more anaerobic ATP production through the activation of a Pparalpha pathway (3).

Flow Cytometry-Anti-PHD1/prolyl hydroxylase antibody [EPR2](TA307021); Overlay histogram showing HeLa cells stained with TA307021 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was DyLight 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monclonal) (1ug/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed.
