|Lipin-1 is a Mg(2)+-dependent phosphatidate phosphatase (PAP) enzyme which catalyzes the conversion of phosphatidate to diacylglycerol, a key reaction in glycerolipid biosynthesis (1). It plays critical roles in controlling energy metabolism, has establishing the adipogenic gene expression program during early stages of differentiation, and in triglyceride accumulation in mature adipocytes (2, 3). Lipin-1 also functions as a transcriptional coactivator in conjunction with members of the peroxisome proliferator-activated receptor (PPAR) family (2). There are two isoforms of Lipin-1 (alpha and beta) which appear to serve distinct functions in adipocytes, with unique expression dynamics, subcellular localization, and effects on gene expression. Lipin-1B is the predominant form in mature adipocytes. Lipin-1 expression levels are associated with insulin sensitivity, metabolic rate, hypertension, and risk for the metabolic syndrome (4). |

Flow Cytometry-Anti-Lipin 1 antibody [EPR3725](TA307006); Overlay histogram showing HepG2 cells stained with TA307006 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22Â°C. The secondary antibody used was DyLight? 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22Â°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1Âµg/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed.
