p16 also known as INK4a and CDKN2A (1-2) is a cyclin-dependent kinase inhibitor that specifically inhibits CDK4 and CDK6. Disruption of cyclin-CDK4 complex by p16 prevents phosphorylation of Rb, therefore preventing cell cycle progression from the G1 to S phase (3-4). p16 suppresses cell growth in response to intracellular stimuli. p16 is also know as an important tumor suppressor. Mutations and deletions in p16 have been detected in a wide variety of tumors, especially in Melanoma (5).
