|MEK1 and MEK2 (MAPK kinase 1/2, or ERK kinase 1/2) are mitogen-activated protein kinases that stimulate MAP kinase activity, playing a role in both cell growth and differentiation (1,2). MEK itself is activated via phosphorylation at serines 217/218 and 221/222 by upstream activator kinases, including c-raf, mos and MEK kinase (3,4). MEK1 and MEK2 are activated by a wide variety of growth factors and cytokines, and also by membrane depolarization and calcium influx (5).|

Immunocytochemistry/ Immunofluorescence - MEK1 antibody [E342]; Ab32091, at a 1/100 dilution, staining MEK1 in A431 cells by Immunofluorescence.
Flow Cytometry - MEK1 antibody [E342]; Overlay histogram showing HeLa cells stained with ab32091 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was DyLight? 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1µg/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed.
