FunctionHistone deacetylase 2 (HDAC2) belongs to the histone deacetylase family type 1 which is responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4) (1). HDAC2 represses transcription and binds transcription factor YY1 to form a large complex (2). It also forms transcriptional repressor complexes by associating with MAD, SIN3, N-CoR and DNMT1 (3-4). Similar to HDAC1 and HDAC3, HDAC2 does not possess a repression domain, but preferentially associates with HDAC1. HDAC2 with HDAC1 is also implicated in a complex of multiproteins that functions through modifying chromatin structure to keep genes silent (5).

Immunohistochemistry (Frozen sections) - HDAC2 antibody [Y461]; TA300044 staining HDAC2 in Rat spinal cord tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with paraformaldehyde and blocked with 1% BSA for 30 minutes at 25°C. Samples were incubated with primary antibody (1/500 in PBS + 0.2% TritonX + 1% BSA) for 16 hours at 4°C. An Alexa Fluor?488-conjugated Donkey anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody. Antigen unmasking with sodium citrate buffer (10mM sodium citrate, 0.05% Tween 20, ph6) was necessary to obtain a good signal. The sections were counterstained with DAPI.

Flow Cytometry-HDAC2 antibody [Y461](TA300044); Overlay histogram showing HeLa cells stained with TA300044 (red line). The cells were fixed with methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was DyLight 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG (1ug/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a decreased signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.
