FunctionRUNX1 (PEBP2-alpha B, Runt-related transcription factor 1, AML1) is a pivotal transcription factor essential for haematopoietic stem cell generation in the vascular regions of the aorta, vitelline and umbilical arteries, yolk sac and placenta (1). It binds DNA in cooperation with CBFbeta to activate or repress transcription, dependent upon cellular context and interaction with a variety of co-activators and co-repressors (2). RUNX1 is required for maturation of megakaryocytes and differentiation of T and B cells (3). It is one of the most frequent targets of chromosome translocations associated with leukemia (4). Mutation at the C-terminal region of RUNX1 might predict acute myeloid leukemia transformation (5).
