|NEK2, a member of the NEK Serine/Threonine protein kinase family, is a protein kinase that is involved in cell cycle regulation (1). It promotes the splitting of duplicated centrosomes during the G2/M transition of mitosis through the phosphorylation of core centriolar proteins (2). Almost undetectable during G1, NEK2 accumulates progressively throughout S phase, reaching maximal levels in late G2 (1). It is structurally related to the nimA kinase (never in mitosis A), which also regulates cell cycle progression (3). During mitotic progression, NEK2 may be functionally similar to nimA in controlling the timing of mitotic entry, chromatin condensation, spindle organization, and cytokinesis (4). Nek2 is ubiquitously expressed in humans as two alternative splice variants, called Nek2A (48 kDa) and Nek2B (44 kDa). Nek2A exclusively phosphorylates and activates NEK11 in G1/S-arrested cells in the nucleolus, while the shorter isoform (Nek2B) is predominantly cytoplasmic (1, 2). Both isoforms are detected in primary blood lymphocytes as well as adult transformed cells (1).|
