BackgroundMinichromosome maintenance protein 2 (MCM2) is a member of the MCM family that regulates mammalian DNA replication (1). This family is composed of six related subunits, called the hexameric MCM2-7 complex, that are conserved in all eukaryotes. It functions as a replicative helicase, the molecular motor that both unwinds duplex DNA and powers fork progression during DNA replication (2). During cell division, MCM2 is required for the entry into S phase (3). It acts as a factor to license DNA for one and only one round of replication per cell cycle (3). MCM2 physically interacts with and is phosphorylated by Cdc7/Dbf4, thus regulating the initiation of DNA replication (4). Ser27, Ser41, and Ser139 are suggested to be Cdc7/Dbf4 phosphorylation sites in MCM2 (5).
