|Cyclin D1 belongs to the Cyclin D family. Cyclin D1 is required for the cell cycle G1/S transition (1). Cyclin D1 forms a serine/threonine kinase holoenzyme complex with and functions as a regulatory subunit of CDK4 or CDK6. Cyclin D1 also phosphorylates and inactivates the retinoblastoma protein and promotes progression through the G1/S phase (2-3). Amplification or overexpression of cyclin D1 plays a pivotal role in the development of various human cancers including parathyroid adenoma, breast cancer, colon cancer, lymphoma, melanoma, and prostate cancer (4-5). Over-expression of cyclin D1 is known to correlate with the early onset of cancer and risk of tumor progression and metastasis (6). Cyclin D1 also associates with, and regulates the activity of transcription factors, coactivators and corepressors that govern histone acetylation and chromatin remodeling proteins (7-8). |

Western blot - Cyclin D1 antibody [ep272y]; Anti-Cyclin D1 antibody [EP272Y] at 1/2000 dilution + MCF-7 cell lysate at 10 µg.Predicted band size : 33 kDa.Observed band size : 36 kDa .
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Cyclin D1 antibody [EP272Y]; Immunohistochemical analysis of paraffin-embedded human kidney carcinoma using anti-Cyclin D1 RabMAb.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Cyclin D1 antibody [EP272Y]; Immunohistochemical analysis of paraffin-embedded human hepatocellular carcinoma using anti-Cyclin D1 RabMAb.
Immunocytochemistry/ Immunofluorescence - Anti-Cyclin D1 antibody [EP272Y]; TA300599 staining cyclin D1 in MCF7 cells treated with KN-93 (water soluble) , by ICC/IF. Decrease in cyclin D1 expression correlates with increased concentration of KN-93 (water soluble), as described in literature. The cells were incubated at 37°C for 24 hours in media containing different concentrations of (KN-93 (water soluble)) in DMSO, fixed with 100% methanol for 5 minutes at -20°C and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with TA300599 (1/100 dilution) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 goat anti-rabbit polyclonal antibody at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.
Immunocytochemistry/ Immunofluorescence - Cyclin D1 antibody [ep272y]; Immunofluorescence staining of Cyclin D1 on MCF7 cells, using TA300599 at a dilution of 1/50.
