BORA is an activator of protein kinase Aurora A, which is required for centrosome maturation, spindle assembly, and asymmetric protein localization during mitosis (1). It is also a Plk1-interacting protein, which controls multiple transitions in mitosis and cytokinesis. One of Plk1's key biochemical functions is to phosphorylate its substrates and to target them for degradation by the SCF-β-TrCP ubiquitin ligase in the cell cycle. Degradation of BORA after mitotic entry releases Plk1 free to associate with prime-phosphorylated substrates, which enhance its kinase activity and target Plk1 to kinetochores and centrosomes for its mitotic function (2).
