|S100A4 is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and they are involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 proteins include at least 13 members. S100A4 may function in motility, invasion, and tubulin polymerization. Altered expression of S100A4 have been implicated in tumor metastasis.|

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-S100A4 antibody [EPR2761(2)]; Immunohistochemical analysis of Human lung tissue, staining S100A4 with TA310834.Tissue was fixed with HOPE and blocked with blocking solution for 5 minutes at 25Â°C. Samples were incubated with primary antibody (1/1000 in diluent) for 1 hour at 25Â°C. An undiluted HRP-conjugated goat anti-rabbit polyclonal IgG was used as the secondary antibody.
Immunocytochemistry/ Immunofluorescence - Anti-S100A4 antibody [EPR2761(2)]; TA310834 staining S100A4 in the A549 cell line from Human lungs by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde and permeabilized with Triton X-100 0.25% in PBS. Samples were incubated with primary antibody (1/100) for45 minutesat 25Â°C. A TRITC-conjugated Goat anti-rabbit polyclonal was used as the secondary antibody.
Flow Cytometry - Anti-S100A4 antibody [EPR2761(2)]; Overlay histogram showing HeLa cells stained with TA310834 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22Â°C. The secondary antibody used was Alexa Fluor? 488 goat anti-rabbit IgG (H+L) at 1/2000 dilution for 30 min at 22Â°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1Âµg/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
