FunctionMembers of the protein kinase C (PKC) family of serine/threonine kinases play a key role in regulating the differentiation and growth of diverse cell types and, to date, the cloning of seven mammalian PKC genes encoding eight distinct isoforms has been reported. PKC gamma (PKCG) is calcium, diacyglycerol, and phorbol esters activated, serine- and threonine-specific enzyme. PKC gamma expression is specific to neurons and it plays an important role in neural signal transduction (1). After activation by phorbol, it moves to cytoplasm, where it shown to regulate intracellular communication in epithelial cells. PKC has three phosphorylation sites in its kinase domain, Thr 514, 655, and 674. Phosphorylations of these three sites are critical for full activation of PKC in response to various stimulants (2). PKC gamma is phosphorylated at Thr 514 by PDK1. PKC gamma is required for long term potentiation (LTP) and long term depression (LTD). Defect in PKC gamma has been linked to neurodegenerative disorder spinocerebellar ataxia-14 (SCA14) (3).
