|The matrix metalloproteinase (MMP) family (approximately 25 members in mammals) has been implicated in extracellular matrix remodeling associated with embryonic development, cancer formation and progression, and various other physiological and pathological events. MMP-14 is a membrane-bound matrix metalloproteinase (MT-MMP) capable of mediating pericellular proteolysis of extracellular matrix components. MMP-14 is therefore thought to be an important molecular tool for cellular remodeling of the surrounding matrix (1). Gelatinase A (type-IV collagenase; M(r) 72,000) is produced by tumour stroma cells and is believed to be crucial for their invasion and metastasis, acting by degrading extracellular matrix macro-molecules such as type IV collagen. MMP-14 may thus trigger invasion by tumor cells by activating pro-gelatinase A on the tumor cell surface (2). Observations identify MT1-MMP as a tumor-derived growth factor that regulates proliferation by controlling cell geometry within the confines of the 3D extracellular matrix (ECM) (3). |

Immunohistochemistry (Paraffin-embedded sections) - MMP14 antibody [EP1264Y]; Ab51074 (1:100) staining human MMP14 in human breast carcinoma tissue by immunohistochemistry using paraffin embedded tissue.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - MMP14 antibody [EP1264Y]; TA300908 at 1/500 staining human kidney tissue sections by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step (in Tris/EDTA) was performed. The tissue was then blocked with serum and incubated with the primary antibody. A biotinylated donkey anti-rabbit IgG was used as the secondary.
Flow Cytometry - MMP14 antibody [EP1264Y]; Overlay histogram showing HT1080 cells stained with TA300908 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody for 30 min at 22°C. The secondary antibody used was DyLight 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG (1ug/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HT1080 cells fixed with methanol (5 min) used under the same conditions.Please note that Abcam does not have data for use of this antibody on non-fixed cells.
