|Bid, BH3 interacting domain death agonist, is a pro-apoptotic member of the Bcl-2 family of proteins that regulate aptosis. Bid is localized to the cytosol in an inactive precursor form. Upon caspase-8 binding in the Fas signaling pathway, Bid is cleaved and the active Bid fragment translocates to mitochondria and induces cytochrome c release and the resulting apoptotic cascade (1-3). Bid contains a BH3 domain which allows it to dimerize with and counter the death repressor effects of Bcl-2. BID has also been shown to heterodimerize with Bcl-x and the death agonist Bax (4).|

Immunohistochemistry (Paraffin-embedded sections) - Bid antibody [Y8]; Immunohistochemical analysis of paraffin-embedded human prostate carcinoma using TA300334 at 1/100 dilution.
Flow Cytometry-Anti-Bid antibody [Y8](TA300334); Overlay histogram showing HeLa cells stained with TA300334 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22Â°C. The secondary antibody used was DyLight? 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22Â°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1Âµg/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line). Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
