HEK293T cells were transfected with the pCMV6-ENTRY control (Cat# PS100001, Left lane) or pCMV6-ENTRY TANK (Cat# RC209759, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-TANK(Cat# TA810317). Positive lysates LY418162 (100ug) and LC418162 (20ug) can be purchased separately from OriGene.

  Western blot analysis of extracts (35ug) from cell lines by using anti-TANK monoclonal antibody (1:500).

• 

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type HeLa cells (WT, Cat# LC810HELA) and TANK-Knockout HeLa cells (KO, Cat# LC810199) were separated by SDS-PAGE and immunoblotted with anti-TANK monoclonal antibody TA810317 (1:500). Then the blotted membrane was stripped and reprobed with anti-HSP90 antibody as a loading control.

• Background: The TRAF (tumor necrosis factor receptor-associated factor) family of proteins associate with and transduce signals from members of the tumor necrosis factor receptor superfamily. The protein encoded by this gene is found in the cytoplasm and can bind to TRAF1, TRAF2, or TRAF3, thereby inhibiting TRAF function by sequestering the TRAFs in a latent state in the cytoplasm. For example, the protein encoded by this gene can block TRAF2 binding to LMP1, the Epstein-Barr virus transforming protein, and inhibit LMP1-mediated NF-kappa-B activation.
