• Recommended Dilution: ELISA: In a Sandwich ELISA (assuming 100 µl/well), a concentration of 8.0-16.0 µg/ml of this antibody will detect at least 500 pg/well of recombinant Rat IL-10 when used in conjunction with Biotinylated antigen affinity purified anti-Rat IL-10 (Cat.-No PP1115B1/PP1115B2) as the detection antibody at a concentration of at least 0.5-1.0 µg/ml.
  Western Blot: To detect Rat IL-10 by Western Blot analysis this antibody can be used at a concentration of 0.5-1.0 μg/ml. When used in conjunction with compatible secondary reagents the detection limit for recombinant Rat IL-10 is 0.5-1.0 ng/lane, under reducing or non-reducing conditions.

• Background: IL-10 is an immunosuppressive cytokine produced by a variety of mammalian cell types including macrophages, monocytes, T cells, B cells and keratinocytes. IL-10 inhibits the expression of proinflammatory cytokines such as IL-1 and TNF-alpha. Like IL-4, IL-10 enhances humoral immune responses and attenuates cell-mediated immune reactions. Human IL-10 is active on murine cells, but murine IL-10 is inactive on human cells.
