Recommended Dilution ELISA (End Point Titre: 1/25000).
THP was bound to the wells of a microtitre plate, and sequential dilutions of antibody were added to the wells. After incubation and washing, alkaline phosphatase conjugated anti-mouse Ig was added to the wells. Colour reaction was developed and scored as intense (4+) to none (-). The titre is defined as the maximum dilution yielding a 4+ reaction.

Immunogen Human Tamm-Horsfall protein.
Specificity Anti-Human Tamm-Horsfall protein (THP) is a monoclonal antibody which reacts with an epitope of the urinary mucoprotein.
This monoclonal antibody has shown good localization to the distal tubule and collecting duct as well as to large deposits of Tamm-Horsfall protein which has clogged peri-pelvic veins.
This antibody has been tested against 21 different Human proteins in Solid Phase Radioimmunoassay and has been shown to be specific for tamm-horsfall protein only. Results of ELISA show no visible reaction with Human albumin or transferrin.
Also no staining of Human tissue other than that noted for kidney sections has been observed.

Reconstitution Method Restore with 1.0 ml of distilled water.
Note: Reconstituted product may have a cloudy appearance, however, this in no way will affect activity.

After reconstitution aliquot and freeze unused portions at -20°C in volumes appropriate for single usage.

Background Tamm-Horsfall Protein (THP) is a glycoprotein of approximately 80 kDa containing up to 25% carbohydrate by weight. It exists as a high molecular weight polymer in urine and approximately 50 mg of the THP are excreted per day by adult humans. THP is produced by the epithelial cells lining the thick ascending loop of Henle and distal convoluted tubules of the kidney. THP is also the primary constituent of renal tubular casts and THP aggregation has been implicated as a factor in the pathogenesis of acute renal failure by causing tubular obstruction. Antibody to THP is present in the serum of patients with pyelonephritis and reflus uropathy.

Note Sterility: This reagent is not sterile, but can be sterilized by filtration if necessary.
To minimize loss of volume during filtration, dilute to final working concentration in the appropriate medium before filtration and filter through a 0.22μ Millipore filter (or equivalent).
