Recombinant human SOX9 protein directly binds to RASSF3 promoter sequence, but not TOP2A promoter sequence. EMSA was performed with biotin-labeled oligonucleotides for TOP2A and RASSF3 with or without recombinant SOX9 (OriGene TP308944). Excess unlabeled oligonucleotides (cold probe) served as controls. Both sequences contain the SOX9 peak center obtained from ChIP-seq experiments. Western blot was performed on the same membrane used for EMSA to detect recombinant human SOX9 protein. • Summary: The protein encoded by this gene recognizes the sequence CCTTGAG along with other members of the HMG-box class DNA-binding proteins. It acts during chondrocyte differentiation and, with steroidogenic factor 1, regulates transcription of the anti-Muellerian hormone (AMH) gene.
