Preparation:HEK293T cells were transiently transfected with MegaTran Transfection Reagent (TT200002) and TrueORF cDNA plasmid. Transfected cells were cultured for 48-72 hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix (Sigma), 1mM PMSF and 1mM Na3VO4), and then centrifuged to clarify the lysate. Recombinant proteins were purified through anti-DDK affinity columns. Protein concentration was measured by BCA kit.
Buffer and Storage:10% glycerol, 100 mM glycine, 25 mM Tris-HCl, pH 7.3. Store at -80C. Avoid repeated freeze-thaw cycles. Stable for 3 months from receipt of products under proper storage and handling conditions.

Summary: The hypoxia inducible factor (HIF) is a transcriptional complex that is involved in oxygen homeostasis. At normal oxygen levels, the alpha subunit of HIF is targeted for degration by prolyl hydroxylation. This gene encodes an enzyme responsible for this post-translational modification. Alternative splicing results in multiple transcript variants. Read-through transcription also exists between this gene and the upstream RAB4B (RAB4B, member RAS oncogene family) gene.
