High transfection efficiency Megatran Figure 1 FACS analysis of GFP expression in CHO-K1 cells transfected with a GFP expression plasmid.  Cells were transfected with 3ug DNA with 9ul transfection reagent. 72 hrs post transfection, CHO-K1 cells were harvest by trypsine and analyzed on FACS Calibur. Y axis indicates number of cells expressing GFP with increasing intensity.  Left panel represents untransfected control.  Transfection efficiency is 95% with MegaTran 1.0 (right panel) and 63% for Lipofectamine2000 (middle panel). 

Transfection of 293F Suspension Cells

  MegaTran Suspension

293F suspension cells were tranfected with different amounts of plasmid coding human-IgG-GPI with MegaTran 1.0 and 293Fectin.  The number of cells expressing surface IgG was analyzed by FACS using anti-human IgG-FITC 72 hours post transfection.
 
In this experiment, MegaTran 1.0 effectively transfected 80% of 293F suspension cells, equivalent to that of 293Fectin.

High transgene protein expression

Combining the high transfection efficiency and low cytotoxicity, MegaTran 1.0 transfected cells produce much higher level of transgene proteins.

Transient transfection with IgG vectors into CHO-K1 cell line. CHO-K1 cells were transfected with plasmids containing two different monoclonal antibody genes.  Day 3 and 6 post transfection, IgG titer in supernatant was measured by Octet.  Note that expression level continues to increase 35-60% between day 3 and day 6 with MegaTran 1.0.

Simple protocol
MegaTran 1.0 works well in media containing antibiotic and anti-mycobacteria agents.  Transfection can be done in media with or without serum.  There is no requirement for media changes.  (Click here to read the application guide).

Manufacturing conditions
MegaTran 1.0 reagents do not contain any protein or animal components.  Each aliquot is packaged using sterile techniques to eliminate any microbial contamination.

Cell spectrum
MegaTran 1.0 has been tested with the following common cell lines: CHO; HEK293; 293F suspension cells; Hela; COS-7; NIH3T3; MEF; U2OS.
