TRADD (TNF-R1 associated death domain) is a multifunctional signaling adaptor protein that is essential in recruiting signaling molecules to the TNFR1 receptor complex (1, 3). It mediates TNFR1 activation of NF-kappaB and c-Jun N-terminal kinase (JNK), as well as caspase-dependent apoptosis (2). The C-terminal of TRADD comprises the "death domain" that is responsible for association of TNFR1 and other death domain-containing proteins such as FADD and RIP. The N-terminal domain promotes the recruitment of TRAF2 to TNFR1, leading to the activation of JNK/AP1 and NF-kappa B (1). TRADD is suggested to also be involved in IFN-gamma signaling by forming a complex with STAT1-alpha within the nucleus and regulating IFN-gamma-mediated STAT1-alpha activation (3).
