Functionp53 acts as both a tumor-suppressor and transcription factor that, upon activation by DNA damage and other cellular stress signals, leads to the transcription of genes triggering cell-cycle arrest, apoptosis, and DNA repair (1,2). The gene for the nuclear phosphoprotein p53 is the most commonly mutated gene yet identified in human cancers (3). p53 is frequently mutated or inactivated in about 60% of cancers. p53 acetylation at K373 is confirmed to be required for recruitment of p300 to the p21 promoter, and the depsipeptide-induced p53 acetylation at K373 is unlikely to be dependent on p53 phosphorylation at Ser15, Ser20, and Ser392 sites. Data suggest that p53 acetylation at K373 plays an important role in depsipeptide-induced p21expression (4,5).

Flow Cytometry - Anti-p53 (acetyl K373) antibody [EP356(2)AY]; Overlay histogram showing HeLa cells stained with TA301070 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was Alexa Fluor? 488 goat anti-rabbit IgG (H+L) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1µg/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.
